Abstract
This study was conducted to investigate the preventive effect of ramie leaf powder (RP) on the loperamide-induced constipation in rats. Male Sprague-Dawley rats were pretreated with RP contained in diets at the concentration of 0, 5 and 10% for 34 days to test the anti-constipation effects. Rats were divided into 6 groups; normal diet group (NOR), normal diet and loperamide treated group (NORL), 5% of heat-dried RP diet and loperamide treated group (HRLL), 10% of heat-dried RP diet and loperamide treated group (HRHL), and 5% of freeze-dried RP diet and loperamide treated group (FRLL), 10% of freeze-dried RP diet and loperamide treated group (FRHL). Constipation was induced by subcutaneous injection of loperamide (1.5 mg/kg body weight/day) for the final 6 days of the experiment. Treatment of loperamide resulted in decreases in the number and wet weight of fecal pellets, and fecal water content. However, the number and wet weight of fecal pellets, and fecal water content were increased in RP-treated groups compared to the loperamide-treated groups. The decreased number and wet weight of fecal pellets, and fecal water content by loperamide were increased by feeding RP, as a dose-dependant manner. Furthermore, gut transit time and transit speed were shortened, and length of gut was elongated in RP plus loperamide compared to loperamide alone. There were no differences in the fecal pellet number, weight, and water contents, and the gut transit time, transit speed, and length between the heat-dried RP group and the freeze-dried RP group. The activities of ALT, AST, ALP and LDL in serum were not significantly different between experimental groups. Serum total cholesterol and LDL-cholesterol concentrations were lowered, and HDL-cholesterol concentration was increased in rats fed the RP diets in proportion to a feeding amount of RP. Serum lipid profiles were not significantly affected by loperamide administration. These results demonstrated that constipation was improved by feeding of RP to rats.
