ABSTRACT

Effect of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf Extracts on Gastric Cancer Cells

Objective : This study aims to discover on the effects of the anti-cancer activity of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf extracts on gastric cancer cells.

Methods : MTS assay is used to detect the effects of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf extracts on the viability of ovarian cancer cells. Western blotting is used to see the effects of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf extracts on inducible nitric oxide synthase(iNOS) and cyclooxygenase-2(COX-2) expression.

Results : Methanol extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf reduce gastric cancer cells depending on time and concentration. Also, the extracts significally reduce iNOS expression at various concentrations.

MTS assay results 1. Methanol extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf reduced ovarian cancer cells depending on time. 2. Methanol extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf reduced ovarian cancer cells depending on concentration. Cell survival rate decreased substantially depending on time and concentration, as it was reduced significantly at 400㎍/㎖. 3. Hot water extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf did not reduce gastric cancer cells at any time. 4. Hot water extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf did not reduce gastric cancer cells at any concentration level. Cell survival rate did not decrease at any time or concentration.

Western blot results 5. Methanol extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf dramatically reduced inducible Nitric Oxide Synthase(iNOS) expression at varoius concentrations. 6. Hot water extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf significally reduced inducible Nitric Oxide Synthase(iNOS) expression at every concentration level and reduced cyclooxygenase-2(COX-2) expression to approx. 80% at 200㎍/㎖.

Conclusion : Methanol extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf significantly reduced 97% of cell viability of human gastric cancer cell lines at 400㎍/㎖ concentration and 99% of iNOS expression at every concentration level. Hot water extracts of Coix lacrymajobi var. mayuen (Rom.Caill.) Stapf dramatically reduced the expression of iNOS at every concentration level. The afore mentioned research result implies that isolation and purification assay is expected to bring out more various clinical effects.
