Effects of the Exocarp of Citrullus lanatus Extracts on Ovarian Cancer Cells

Objective : This study is on the effects of the anti-cancer activity of extracts from watermelon peels on ovarian cancer cells.
Methods : MTS assay was used to detect the effects of extracts from watermelon peels on the viability of ovarian cancer cells. Western blotting was used to see the effects of extracts from watermelon peels on inducible nitric oxide synthase(iNOS) and cyclooxygenase-2(COX-2) expressions.
Results : Methanol extracts from watermelon peels slightly reduced ovarian cancer cells depending on time and concentration. Also it dramatically reduced cyclooxygenase-2(COX-2) and iNOS expressions at various concentrations.
MTS assay results are as follows ;
1. Methanol extracts from watermelon peels slightly reduced ovarian cancer cells depending on time.
2. Methanol extracts from watermelon peels slightly reduced ovarian cancer cells depending concentration. There are no difference in cell survival rate at any time or concentration.
3. Hot water extracts from watermelon peels did not reduce ovarian cancer cells at any given time.
4. Hot water extracts from watermelon peels did not reduce ovarian cancer cells at any concentration. There was no difference in cell survival rate at any given time or concentration.
Western blot results are as follows ;
5. Methanol extracts from watermelon peels dramatically reduced inducible Nitric Oxide Synthase(iNOS) expression at various concentration and reduced cyclooxygenase-2(COX-2) expression at the concentration of 100~200㎍/㎖.
6. Hot water extracts from watermelon peels dramatically reduced inducible Nitric Oxide Synthase(iNOS) expression at various concentration and reduced cyclooxygenase-2(COX-2) expression at 200㎍/㎖.
Conclusion : Methanol extracts of watermelon peels slightly reduced cell viability of human ovarian cancer cell lines by 10-20% at 400㎍/㎖. Also, Methanol extracts reduced iNOS expression by 92% and COX-2 expression at various concentration. Hot water extracts of watermelon peels reduced the expression of iNOS at all concentrations.
The effects of Methanol extracts of watermelon peels on the reduction of cell viability suggest that implementing isolation and purification to the tests can bring out more various clinical effects.
