Abstract
Antibacterial compound produced by Bacillus subtilis MJP1 was purified. The cell-free culture supernatants from B. subtilis MJP1 were seperated using C18 Sep-Pak cartridge, ion exchange chromatography(IEC), and gel filtration chromatography(GFC). The purified antibacterial compound showed antibacterial activity against Bacillus subtilis ATCC 6633, Listeria monocytogenes ATCC 19113, Staphylococcus aureus subsp. aureus ATCC 29213, Enterococcus faecalis ATCC 29212. The purified antibacterial compound was found to be stable in the pH range of 3.0∼9.0, but it was unstable at pH 10.0 Also, the antibacterial compound was stable at 50℃ for 24 h, and after lipase and α-amylase treatment. However it was inactivated by proteinase K, pronase E, and heat treatment at 121℃ for 15 min, which indicated its proteineous nature. Ultra performance liquid chromatography(UPLC) and electrospray ionization tandem mass spectrometry(ESI-MS/MS) analysis were used to identify the purified antibacterial compound and confirmed the existence of two peptides(A:3356.54 Da, B:3400.52 Da).
