Abstract
Purpose: Cytokines involve in both inflammatory and healing mechanism. The authors wanted to evaluate the changing nature of cytokines and thereby appropriate time for using bone marrow derived monuclear cells(BM-MNC) to acquire best effect.
Materials and Methods: Bone marrow was aspirate from patients who consented to the procedure. Cytokines from plasma layer acquired from 30 minutes of centrifugation at 1,820 rpm was analyzed. Cytokines were evaluated after abstraction of mononuclear cells using ficoll method. The cytokines evaluated were Eotaxin, GM-CSF, IFN r, IL-1 a, IL-1 b IL-10, IL-12(p40), IL-12(p70), IL-13, IL-15, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IP-10, MCP-1, MIP-1a, RANTES, TNF-a.
Results: High level of IP-10, IL-12 p40, RANTES were detected after centrifugation. Additionally, high level of IL-6, IL-7, IL-8, MCP-1, MIP-1a were noticed 1, 7days following culture.
Conclusion: Mononuclear cells derived from bone marrow has high activity character and change in cytokine was observed after simple culture. Our results show that bone marrow derived mononuclear cells immediately after centrifugation of bone marrow is most effective.
