Abstract
This study was performed to determine the anti-cancer effects of porphyran-contained Chungkookjang, because Chungkookjang has been reported to stimulate cancer cell death. Porphyran is a major component of the red algae of the genera, Porphyra tenera and P. yezoensis from the Porphyra species. Porphyran is one of the sulfated polygalactans composed of galactose and 3,6-anhydrogalactose unit. Crude porphyran was efficiently extracted with 50 vol.(w/v) of 0.1 N HCl at 60℃ for 3 hours. The chemical properties of porphyran from waste laver (Porphyra yezoensis) were measured. The yields of crude porphyran was 13.9%. Contents of total sugar, sulfate/total sugar, protein and 3,6-anhydrogalactose in the crude porphyran were 66.6%, 16.9%, 8.6%, and 16.1% respectively. The effects of porphyran on cytotoxicity to human normal cell line (BJ) and human cancer cell lines (AGS and HT-29) were examined. Porphyran caused a significant decrease in viability in both gastric cancer cells and colon cancer cells, however, did not affect the viability of normal cells. Chungkookjang a kind of fermented soybean pastes representing Korea's fermented food using soybeans has been known to be abundant source of proteins and lipids with digestability as fermented seasoning food and to have strong antimutagenic and cytotoxic effect on cancer cells. Chungkookjang was prepared by inoculating Bacillus subtilis into steamed soybeans at 1%(v/w) concentration and fermenting at 37℃. Physio-chemical properties and in vitro anticancer effects of Chungookjang which has porphyran or not were investigated. The changes in temperature, total amino acid and free amino acid compositions, and average hydrophobicity during fermentation of the Chungkookjang were measured. The cytotoxicity effects of methanol extracts and water extracts from Chungkookjang or Chungkookjang combined with porphyran were evaluated with MTT assay using human foreskin normal cell(BJ) and human cancer cells (AGS, HT-29). The anticancer effect of methanol extracts from both Chungkookjang and porphyran containing Chungkookjang was more stronger than that of water extracts from same sources. All extracts did not affect the growth of normal cells. However, the methanol extract of Chungkookjang combined with porphryan showed higher anticancer effects than that of Chungkookjang in both AGS and HT-29 cells.
