Abstract
The purpose of this study was to evaluate the level of MMPs such as MMP-1, MMP-8, and MMP-13 and TIMP-1 expressed by human periodontal ligament fibroblasts (hPDLs) via western blotting method.
hPDLs were obtained from a healthy periodontium and cultured in Dulbecco's modified Eagles's medium plus 10% fetal bone serum. The concentration of tertacycline-HCl (TC-HCl) application in hPDLs was 250㎎/5㎖, 250㎎/100㎖, 250㎎/1,000㎖, 25㎎/3,000㎖, 250㎎/5,000㎖, 250㎎/7,000㎖, 250㎎/9,000㎖, and 250㎎/10,000㎖. TC-HCl was applied with hPDLs for 1-min. a day over 5 days, respectively. The gene expression on the proenzyme forms and/or enzyme forms of MMP-1-, MMP-8-, MMP-13-, and TIMP-1-mRNA in hPDLs was analysed at 6-day after TC-HCl application via western blotting method. Within the limited experiments, the present data showed that both the protein expression of MMPs (especially MMP-13) and TIMP-1 was increased after 250㎎/5㎖ of TC-HCl application in hPDLs. In future, more research will be needed to evaluate the effect of increased MMPs and TIMP-1 on periodontal cells
