Abstract
Purpose : We explored the relationship between the expression of p53 and γ-H2AX gene in testicular germ cells of rats with experimental left varicocele (ELV) and apoptosis of germ cells.
Materials and method : A total of 16 adult male Sprague-Dawley rats were studied in 2 groups, namely group 1-12 with varicocele, group 13-16 with sham operation. The varicocele-induced rats underwent orchiectomy after 2, 3, 4 weeks and the sham operation rats underwent orchiectomy after 4 weeks. The sections of the left testis from each animal were studied. p53 and γ-H2AX gene expression were determined with immunohistochemistry and western blot.
Results : The p53 and γ-H2AX gene expression were began immunohistochemically at two weeks, remarkable at 3, 4 weeks. Although p53 expression in western blot was not clear at two weeks, but it was considerable at 3, 4 weeks compared with control. The γ-H2AX expression in western blot was considerable at two weeks, remarkable at 3, 4 weeks compared with control.
Conclusions : In the varicocele of rats increased expressions of p53 and γ-H2AX gene may faciliate apoptosis in the rat testis.
