Pig27 및 Pn23 DNA 프로브를 이용한 Prevotella intermedia 및 Prevotella nigrescens의 검출

- Abstract
- The purpose of this study is to develop species-specific DNA probe for the identification of Prevotella intermedia and P. nigrescens. The HindⅢ-digested genomic DNAs of P. intermedia ATCC 49046 and P. nigrescens ATCC 33563^(T) were cloned by shotgun method. The candidate species-specific DNA fragments were primary screened by inverted dot blot hybridization and then confirmed by Southern blot analysis. The nucleotide sequences of species-specific DNA probes were determined by chain termination method. The data showed that Pig27 and Pn23 DNA probes were specific to the P. intermedia and P. nigrescens, respectively. The Pig27 DNA probe contains leucine rich repeat domain protein and ADP-ribosyglycohydrolase. These results indicate that the Pig27 and Pn23 DNA probes could be useful in detection of the P. intermedia and P. nigrescens, respectively, at the species level.
