두경부 암세포에 대한 capsaicin의 항암효과

- 음식물 첨가물로 이용되고 있는 고추의 매운 맛 성분인 capsaicin (trans-8-methyl-6-nonenamide)을 두경부 암세포인 FaDu 세포에 처리하여 capsaicin이 암세포의 apoptosis을 유발하여 항암효과를 갖는 지를 세포 및 유전자 수준에서 평가하고자 cell proliferation assay (MTT), cytochrome C ELISA assay, caspase ―3 enzyme activity assay, fluorescent activated cell sorting (FACS) analysis에 의한 apoptosis 관찰, DNA fragmentation assay, protein kinase C (PKC) 분석 실험들을 수행하였다. Capsaicin을 처리한 FaDu 세포들은 염색질(chromatin)의 응축, endonuclease의 활성화에 의한 DNA 규칙적인 절단 즉, 사다리 모양의 분절을 나타냈으며 세포질에서 cytochrom C가 방출 되었고 capsaicin에 처리 된 FaDu 세포에서 caspase―3의 활성 증가 및 caspase―3억제제인 Ac―DEVD―CHO를 처리 시 caspase―3 의 활성을 억제하였다. 이러한 결과들은 두경부암 치료와 예방에 적용 될 수 있는 capsaicin의 응용 가능성과 capsaicin을 이용한 식이 요법 개발 가능성을 제시한다.|Capsaicin (trans―8―methyl―6―nonenamide), a spicy component of hot pepper, is a homovanillic acid derivative. Several recent reports indicate that capsaicin could be beneficial on certain types of cancer including colon cancer. In this study, for the evaluation of chemopreventive activity of capsaicin on pharyngeal cancer, we investigated whether capsaicin induces apoptotic cell death in FaDu human pharyngeal squamous carcinoma cells. Capsaicin inhibited growth of FaDu human pharyngeal squamous carcinoma cells in a dose dependent manner. Apoptotic cell death was confirmed by the analyses of DNA fragmentation, nuclear condensation and propidium iodide (PI) staining with fluorescent activated cell sorting (FACS). The increase in the cytosolic cytochrome C level and the activation of caspase 3, in response to the capsaicin treatment, were confirmed by cytochrome C assay and caspase 3 cellular activity assay, suggesting that apoptosis was induced by cytochrome C triggered caspase 3 activation. Current data suggest that capsaicin may have beneficial effects for the treatment of pharyngeal cancer.
