한국인 교정환자의 치면세균막으로부터 Streptococcus downei의 분리

Abstract

최근 뮤탄스 연쇄상구균을 선택적으로 배양할 수 있는 mitis-salviarius bacitracin (MSB) aga 배지에서 제Ⅰ형 또는 제Ⅱ형의 생물형을 갖는 4 균주를 분리하였다. 본 연구에서는 이들을 분자생물학적 방법을 이용하여 종 수준에서의 동정을 위해 시행하였다. 이들 4 가지 임상균주들의 지놈 DNA를 추출하고, 16S rDNA를 중합효소연쇄반응법을 이용하여 증폭한 다음 클로닝한 다음 핵산염기서열을 결정하였다. 연구결과 4개의 임상균주들의 16S rDNA 핵산염기서열이 기존에 알려진 Streptococcus downei ATCC 33748^(T)균주의 것과 99% 이상 상동성을 갖는다는 알 수 있었다. 사람에서 Streptococcus downei 균종이 분리 동정되었다는 보고는 아직까지 미국립보건원에서 제공하는 PubMed 데이터베이스에는 찾을 수가 없다. 이상의 연구 결과를 종합할 때, 아직까지 사람의 치면세균막에서의 존재 유무가 명확하게 알려지지 않은 S. downei 균종이 사람에게도 존재함을 알 수 있었으며, 차후 이들의 특성을 밝히는 실험과 다른 국가나 인종에도 존재하는 지를 국제연구를 통해 진행한다면 좋은 결과를 얻을 수 있을 것으로 사료된다.|Recently, we isolated 4 bacterial strains growing on the MSB agar that have the similar biochemical characteristics with biotype Ⅰ or Ⅱ. The aim of this study was to identify the 4 clinical isolates. The bacteria were identified using 16S rDNA cloning and sequencing methods. The results showed that the 16S rDNA sequences of 4 clinical isolates had above 99 % similarity with that of Streptococcus downei ATCC 33748^(T). To our knowledge, it is the first report that S. downei are isolated from dental plaque in human. These results indicate that S. downei could also inhabit dental plaque of human.
