Kidney had recovery function toxicants, ischemia, reperfusion -induced damage, acute-renal failure (below, called ARF).
Fibroblast growth factors (below, called FGF) constitute a 7large family of signalling molecules that participate in regulation of basic cellular processes such as proliferation, survival, migration and differentiation. Basic fibroblast growth factor (below, called bFGF) is an angiogenic growth factor that is involved the pathogenesis of renal diseases. bFGF was released into the renal tissue by injured endothelial and renal epithelial cells and was a one of the important marker of renal disease including chronic renal failure, acute renal failure.
Squalene (below, called SQ) is a triterpenoid hydrocarbon oil, C30H50 (2,6,10,15, 19, 23-hexamethyl-2,6,10,14,18,22-tetracosahexene) that have antioxidant effect, detoxic effect against diverse chemicals, protective effect against radiation and anti-cancer effect.
The aim of this study is to evaluate the effects squalene on the prevention of experimental acute renal failure (below, called ARF) induced by glycerol.
In case of in vitro study, we were investigated the expression of bFGF by RT-PCR. After the proximal tubular cells was isolated, added to glycerol (1,2,4 mM) or plus squalene (0.1, 0.05 or 0.1 %). In case of in vivo study, we were investigate the changes of BUN, creatine and expression of bFGF mRNA and protein. The experimental groups were divided into three groups. Group 1 was normal mouse. Group 2 was not treatment with squalene after intraperitoneal contamination of glycerol (50 %, 8 ml/kg). And, Group 3 was treated with squalene (180 ㎎/㎏) after intraperitoneal contamination of glycerol (50 %, 8 ml/kg). All groups were used to 7 mice.
In the results, we investigated that glycerol induced the renal failure. And, the expression of bFGF mRNA was increased in renal PTC. But, SQ showed that the expression of bFGF mRNA was decreased in renal PTC. And, SQ was quickly recovered the levels of BUN and creatine which compared with treated only glycerol (P<0.01).
It was concluded that SQ effected to the glycerol induced-renal disease and may be related to the regeneration by bFGF in the renal PTC.
