- Abstract
- The classical (TRPC) subfamily of transient receptor potential (TRP) channels are regarded as receptor-operated Ca^(2+)-permeable cation channels activated in a phospholipase C (PLC)-dependent pathway. TRPC channels can be divided into four subgroups according to sequence similarity. Group 4 is composed of TRPC4 and TRPC5, and is most closely related to group 1 (TRPC1). The distribution and functional properties of TRPC4-immunoreactivity(IR) in mammalian intestine are not well-known. In this study, therefore, we examined the distribution of the TRPC4 in the murine small intestininal wall, with emphasis on interstitial cells of Cajal(ICC). We observed the TRPC4-IR in the cultured smooth muscle cell and ICC using double immunolabelling. The distribution of TRPC4-IR in the whole-mount preparation was also revealed in the myenteric plexuses, the smooth muscle cells and the ICC. Double-labelling showed that c-kit-IR ICC in the circular muscle layer and myenteric plexus expressed TRPC4. This datas showed that TRPC4-IR is abundantly present in the murine small bowel, and subpopulations of c-kit-IR ICC were TRPC4-IR. It is concluded that TRPC4 may play a role in the murine intestinal motility.
