망막색소상피세포에서 산화스트레스에 의한 MMPs의 발현 변화

- Purpose : To evaluate the expression pattern of matrix metalloproteinases(MMPs) in the human retinal pigment epithelial(HRPE) cells by oxidative stress.
Material and Method : ARPE-19 cells were exposed to paraquat for 72 hours. The cells were assayed for the mRNA expression of MMP-2 and MMP-9 by reverse transcriptase polymerase chain reaction(RT-PCR) method. The conditioned media were assayed for the production of MMPs protein by Gelatin zymography and Western blotting. Also, the inhibition effect of antioxidants against the oxidative stress of the cells was checked.
Result : MMP-2 and MMP-9 levels in the RPE cells were increased under the condition of oxidative stress compaired with control (p〈 0.05). Also, the levels were decreased when the cells incubated with an antioxidant, N-acetylcysteine (NAC).
Conclusion : Oxidative stress upregulates MMP-2 and MMP-9 expression in the HRPE cells
