Polo-like kinase 1 phosphorylates HSF1 and mediates its nuclear translocation during heat stress

Abstract
Heat shock transcription factor 1 (HSF1)은 다양한 pathophysiologic stress에 의해 활성화되고, heat shock protein (HSP)의 발현을 증가시킨다. HSF1의 활성은 과인산화와 같은 다양한 스트레스 관련 과정을 통해 발생하지만, 정확한 HSF1 활성 mechanism은 밝혀져 있지 않다. 본 연구자는 Tandem Affinity Purification (TAP) system을 이용하여 polo like kinase 1 (PLK1)과 HSF1이 세포내에서 결합하는 것을 확인했다. HSF1과 PLK1의 결합은 열 스트레스에 의해 증가하지만 PLK1의 과발현은 HSF1의 trimerization과 DNA binding activity에 영향을 주지 않았다. 그러나 PLK1은 HSF1의 419번 아미노산인 serine의 인산화를 유도하였으며, 흥미롭게도 HSF1의 419을 alanine으로 point mutation시키면 HSF1의 핵으로 전이가 저해 받았다. 본 연구의 결과에서 PLK1에 의한 HSF1의 인산화는 HSF1의 핵으로의 전이에 중요한 과정임을 확인하였다.|Heat shock transcription factor 1 (HSF1) is activated by pathophysiologic stresses and activation leads to an increased cellular level of heat shock proteins (HSPs). Although the activation of HSF1 occurs via multiple stress-induced processes such as hyperphosphorylation, the exact cellular mechanism of HSF1 activation is still unclear. Here we showed the interaction of polo-like kinase1 (PLK1) and HSF1 in vivo using the tandem affinity purification (TAP) system. Although the interaction between HSF1 and PLK1 was increased by thermal stress, overexpression of PLK1 did not affect HSF1 trimerization or DNA binding activity. This interaction has resulted in the phosporylation of HSF1 on serine 419 by PLK1. Interestingly, mutation of serine 419 to alanine inhibited heat-stress induced HSF1 nuclear translocation.
Our results suggest that the phosphorylation of HSF1 by PLK1 is an essential step for HSF1 nuclear translocation by heat stress.
