Inhibitory effects of Cafestol and Kahweol on tumor invasion and metastasis

Abstract
본 연구에서는 커피열매에 함유되어 있는 Cafestol(CA)와 Kahweol (KA)의 종양 침투와 전이에 대한 영향을 조사하였다.CA와 KA는 HT-1080fibrosarcoma세포의 이동과 침투를 처리-농도에 의존적으로 억제 시켰다.또한 MMPs의 활성도를 측정하기 위해 Zymography를 한결과 CA와 KA가 MMP-2및 MMP-9의 활성도를 감소시키는 것을 확인할 수 있었다.Endopeptidases의 발현과 종양 전이에서의 단백질 분해 저해에 관련 있는 것으로 잘 알려진 MMPs와 MT1-MMP에 대한 CA와 KA의 영향을 조사한 결과, CA와 KA가 MMP-2, MMP-9과 MT1-MMP의 mRNA 발현을 감소시키고 TIMP-1과 TIMP-2의 발현은 증가 시켰다. Luciferase report assay 결과 MMP-2, MMP-9 의 Promoter가 CA와 KA의 농도에 의존적으로 감소함을 알 수 있었다. MMP-2,MMP-9의 전사조절에 관여하는 것으로 잘 알려진 전사 조절인자 AP-1과 NF-κB 활성에 대한 CA 와 KA의 영향을 조사하기 위해 EMSA와 luciferase report assay를 수행한 결과로부터 CA 와 KA가 AP-1과 NF-κB의 활성을 억제함으로써 MMP-2,MMP-9의 발현을 저해함을 알 수 있었다.실험동물을 이용한 B16F10흑색종 세포의 폐전이 실험결과 CA와 KA가 처리 농도에 의존적으로 B16F10흑색종의 폐 전이가 현저하게 억제되는 현상을 관찰 하였다.결론적으로,CA와 KA의 항전이 활성은 HT-1080세포의 이동억제와 MMP-2,MMP-9발현 저해를 통한 암세포의 침투를 저해함으로써 생긴 결과로 사료되며,본 연구를 통하여 CA와 KA이 암세포에 대한 항전이 효과가 뛰어나다는 사실을 알 수가 있었다.|Cafestol (CA) and its dehydro derivative, Kahweol (KA), are two diterpenes that are present in considerable quantities in coffee beans. The present study investigated the effects of CA and KA on tumor invasion and metastasis. CA and KA inhibited cell migration and invasion of HT-1080 fibrosarcoma cells in a dose-dependent manner. Proteolytic degradation of the tumor metastasis correlate with expression of endopeptidases known as matrix metalloproteinases (MMPs) and membrane-type 1 matrix metalloproteinase (MT1-MMP). The activities of MMP-2 and MMP-9 were inhibited by CA and KA as demonstrated by zymographic assay. Next, the amounts of MMP-9 and MMP-2 protein were analyzed in the cells. MMP-2, MMP-9, and MT1-MMP mRNA expression were suppressed by CA and KA with time and dose-dependent; however, tissue inhibitor of metalloproteinase-1 (TIMP-1) and TIMP-2 expression were increased by CA and KA. Furthermore, effects of CA and KA on the transcription factor binding to MMP-2 and MMP-9 promoter regions in the nucleus were studied using electrophoretic mobility shift assay (EMSA). The transcription factor binding activities of activator protein-1 (AP-1) and nuclear factor κB (NF-κB) were down-regulated by CA and KA. It was found that a decreased transcriptional activity of AP-1 and NF-κB sites by CA and KA were sufficient to reduce MMP-2 and MMP-9 promoter activity. In an in vivo B16F10 melanoma cells lung metastasis experiment, CA and KA showed remarkable inhibitory effects on the lung tumor colonization in a dose-dependent manner. These results demonstrate that anti-metastatic activities of CA and KA resulted from blocking of migration and invasion of the HT-1080 cells. Taken together, the results of this studies provide evidence that CA and KA possess an anti-metastatic activity.
