Development of PCR primers for the detection of Actinobacillus actinomycetemcomitans

Abstract

본 연구는 16S 라이보솜 RNA 유전자의 종간 다양한 핵산염기서열을 보이는 부위를 이용하여 Actinobacillus actinomycetemcomitans를 종-수준에서 동정 및 검출할 수 있는 중합효소연쇄반응 프라이머를 개발하기 위하여 시행되었다. 기존에 알려진 A. actinomycetemcomitans 균주들의 16S 라이보솜 RNA 유전자 염기서열을 이용하여 종-특이 프라이머(ChDC-Aa-F와 ChDC-Aa-R)를 컴퓨터 프로그램으로 설계하고 이를 제작하였다. 프라이머의 특이도는 A. actinomycetemcomitans 9균주, 11균주(3종)의 Haemophilus 속, 그리고, 6균주의 다른 구강 균종을 대상으로 실시하였다. 프라이머의 민감도는 표준균주인 A. actinomycetemcomitans ATCC 33384T의 지놈 DNA를 추출하여 실시하였다. 프라이머의 특이도 실험결과 9균주의 A. actinomycetemcomitans 지놈 DNA에서만 종-특이 중합효소연쇄반응 산물이 증폭되었고, 다른 균종의 지놈 DNA에서는 증폭되지 않았다. 또한 감수성 실험 결과 본 연구에서 사용된 프라이머는 A. actinomycetemcomitans ATCC 33384T 지놈 DNA의 4 fg까지 검출할 수 있었다. 이상의 결과를 종합할 때 ChDC-Aa-F와 ChDC-Aa-R 프라이머들은 A. actinomycetemcomitans 균주를 높은 민감도를 가지고 종-특이적으로 동정 및 검출하는 데 이용될 수 있을 것으로 생각된다. 특히, 치주질환과 A. actinomycetemcomitans간의 역학관계, 치주 치료 전후의 A. actinomycetemcomitans 균주 모니터링 등에 이용될 수 있을 것으로 생각된다.|This study was undertaken to develop PCR primers for the identification and detection of Actinobacillus actinomycetemcomitans using species-specific forward and reverse primers. These primers target variable regions of the 16S ribosomal RNA coding gene (rDNA). Primer specificity was tested against nine strains of A. actinomycetemcomitans, three species (11 strains) of genus Haemophilus and six different species of oral bacteria. The primer sensitivity was determined by testing serial dilutions of purified genomic DNAs of A. actinomycetemcomitans ATCC 33384T. The data showed that species-specific amplicons were obtained for all of A. actinomycetemcomitans strains tested and did not occur with other species. The PCR could detect as little as 4 fg of chromosomal DNA of A. actinomycetemcomitans. These findings suggest that the PCR primers are highly sensitive and are suitable for the applications in epidemiological studies, diagnosis, and monitoring of periodontal pathogens after the treatment of periodontitis.
