Abstract
The aim of this study was to determine if the 5'dUTPase and zinc protease genes are useful in identifying Fusobacterium nucleatum at the subspecies level using a nucleotide sequencing method. Seventeen F. nucleatum strains isolated from the dental plaque of Korean subjects were examined. The 16S rRNA coding gene (16S rDNA), 5'dUTPase and Zinc protease genes were amplified by a polymerase chain reaction and were directly cloned into the pGEM-T easy vector. The nucleotide sequencing was performed using the dideoxy chain termination method, which. identified 17 F. nucleatum strains; 1 strain of F. nucleatum subsp. nucleatum, 1 strain of? F. nucleatum subsp. vincentii/fusiform, 14 strains of F. nucleatum subsp. polymorphum, and 1 unidentified strain (F. nucleatum ChDC F47) at the subspecies level by the nucleotide sequence of 16S rDNA. However, F. nucleatum ChDC F47 was identified as F. nucleatum subsp. vincentii/fusiform from the nucleotide sequences of the 5'dUTPase and zinc protease genes. The results of the identification of the other strains coincided with the nucleotide sequence of the three genes. This suggests that the nucleotide sequencing of 5'dUTPase and zinc protease genes might be useful for identifying F. nucleatum at the subspecies level.
