완도 해조상 및 대형 갈조식물 원형질체(protoplast)배양에 관한 연구

- Abstract
- In this study, I investigated the diversity of seaweeds and their changes in the coastal rocky area of Jeongdo-ri, Wando-Gun, Korea from 2019 to 2020. 135 species were identified based on morphological and molecular approaches. Sheet forms, filamentous forms and coarsely branched forms, which are ESG (ecological state group) II, were more than 60% in all seasons using Litter and Litter (1984) and Orfanidis et al. (2001). Using a 50 × 50 cm2 quadrat method, it was identified that seasonal dominant species (highest value of IV) in upper, middle and lower areas of intertidal zone are Gloiopeltis furcata, Monostroma nitidum and Chondracanthus tenellus in winter; Ulva australis, Corallina pilulifera and Corallina pilulifera in spring; Ulva australis, Sargassum fusiforme and Chondracanthus tenellus in summer; Chondracanthus intermedius, Chondria crassicaulis, Chondracanthus tenellus in fall.
Ecklonia cava, a large brown alga, is in the spotlight as a major species that creates artificial sea forests in coastal ecosystem restoration programs and is mentioned as a candidate for drug development using extracts such as fucoidan and phlorotanin for liver, heart, nerve protection, and immunity. Various studies are being conducted on compound production and modification using E. cava. Recently, protoplast culture has been used as a method for cultivating seaweed in large quantities. This study was planned to develop a new aquaculture method by mixing enzymes and chelate pretreatment agents to find optimal conditions for separating protoplasts from sporophytes of E. cava. Through this study, I determined the optimal conditions for protoplast preparation such as mixing enzymes, chelate pretreatment, pH / time / temperature for incubation, osmotic pressure, proper thallus fragment size, etc., which are the main factors of protoplast separation, were derived. The results of this study are expected to be used in aquaculture to produce E. cava, that would create artificial marine forests, and secure biological quantities.
