Abstract
Analysis of anti-oxidant, anti-aging, and anti-microbial activities of Saccharina japonica stipe and molecular docking analysis of tyrosinase Cho, Jung Hwa Advisor : Prof. Kim, Seong Jae, M.D., Ph.D. Department of Complementary and Alternative Medicine, Graduate School of Chosun University Background: Skin aging is influenced by the activity of enzymes such as tyrosinase. Saccharina japonica is a seaweed rich in antioxidant, anti-aging, and antibacterial components, and it is expected to interact with tyrosinase. Methods: Saccharina japonica stem prethanol extract was prepared and its components and antioxidant activity were analyzed. In addition, tyrosinase enzyme activity inhibition, DOPA oxidation inhibition, hyaluronidase, elastase, MMP-1,12 activity inhibition rates were measured. And, the antibacterial activity against six bacteria including E. coli and Pseudomonas aeruginosa was evaluated. Finally, in silico molecular docking studies were performed with the major components of Saccharina japonica stem prethanol as ligands and tyrosinase as a target. Results: Saccharina japonica stem prethanol had high contents of polyphenol, flavonoid, and water-soluble vitamin C, and showed excellent DPPH radical scavenging ability. It also inhibited tyrosinase activity in a concentration-dependent manner and showed higher DOPA oxidation inhibition than arbutin. Moreover, it significantly inhibited the activity of hyaluronidase, elastase, MMP-1,12, which are involved in the elasticity and collagen synthesis of skin and muscle. The antibacterial activity was comparable to that of vancomycin and imipenem. In the molecular docking study, erucic acid showed the highest binding affinity with tyrosinase and interacted with several amino acids of the tyrosinase protein. Conclusion: This study suggested the possibility of developing Saccharina japonica stem prethanol extract as an excellent natural resource and anti-aging material that can regulate various factors and activities involved in skin aging.
