Abstract
This study was performed to evaluate the therapeutic effect of an oil blend (HC-HS & HS) containing a mixture of Helichrysum (HC) and Hempseed (HS) oil on atopic dermatitis (AD)-induced NC/Nga mice. For this purpose, the skin score, scratching behavior, oil cytotoxicity, superoxide anion-radical (SAR) scavenging ability-dependent anti-oxidant effect, nitric oxide inhibitory ability-dependent anti-inflammatory effect , changes in serum IgE levels, and density of collagen and tumor necrosis factor ((TNF)-α) were assessed in NC/Nga (AD)-induced mice. Additionally, AD-induced mouse tissue was morphologically observed using a light microscope. The skin scores and scratching behavioral indices of the HS, HC, or HH (HC+HS) oil-treated groups, and especially of the HH oil-treated group were more significantly reduced than those of the AD-induced group. There were no significant differences in the cell viability observed between the control and 2% or 3% oil (HC, HS, HH)-treated groups; therefore, none of the oils showed any statistically significant cytotoxic effect. The anti-oxidant and anti-inflammatory abilities of the oil-treated group (HC, HS, HH), and especially the HH oil-treated group were more significantly increased than those in the AD-induced group. During the quantitative measurement of IgE levels, the densities of collagen and TNF-α in the oil (HC, HS, HH)-treated group, and especially in HH oil-treated group were significantly decreased, as compared to those in the AD-induced group. During the light microscopic observation of tissues from AD-induced mice, the oil-treated group (HS, HC, HH) showed a significant decrease in the total levels of collagen and expression of TNF-α, as compared with those in the AD-induced group. Hence, the blended oil-treated group (HH) showed more useful therapeutic effects than the oil (HC, HS)-treated group with regard to the anti-oxidant effect, anti-inflammatory effect, IgE level, total densities of collagen and TNF-α, skin score, and scratching behavior in AD-induced mice. Therefore, it is suggested that essential oils such as HS or HC might be useful for the prevention or treatment of AD. Furthermore, blended oil (HH) is more effective than the each of the oils (HC, HS), because of the synergic effects in AD treatment. Conclusively, oil blends such as HH oil might play an important role in the improvement of AD via aromatherapy.
