Antioxidative, Antihistamine and Antiinflammatory Effect of Chamaecyparis Obtusa Oil by Supercritical Carbon Dioxide Extraction and German Chamomile Oil

Atopic dermatitis (AD) is one of the most common allergic diseases along with allergic rhinitis and bronchial asthma. The of AD patients rapidly increases every year, and its prevalence especially among children and adolescents has been steadily increasing. AD is considered as rather severe and intractable condition.
The cause and mechanisms of AD have not been clearly identified yet. AD is being treated by using antihistamine, an antibiotic drug to prevent inflammation and infection, and steroid, an immunosuppressive and antiinflammatory agent. However, its treatment is difficult because there are various mechanisms of AD and the side effects caused by drugs cannot be neglected.
Recently, some research suggest that aromatherapy using oils extracted from plants with antioxidant effects is effective in treating AD, and it has been gaining much attention for the medical community that such aromatherapy can be a form of medicine for AD treatment. Therefore, this study intends to verify the Antioxidative, Antihistamine and Antiinflammatory effects on AD of the Chamaecyparis obtusa oil extracted through supercritical carbon dioxide extraction process and German chamomile oil.
As for the research method, RBL-2H3 cells, a mast cell line, were treated with supercritical carbon dioxide extracted Chamaecyparis obtusa oil (C oil), German chamomile oil (G oil) and the mixed oil of Chamaecyparis obtusa and German chamomile (CG oil) to perform the electron donating ability (EDA) test, the superoxide dismutase (SOD) like activity test, the lipid peroxidation activity (LPA) analysis, the analysis on their inhibitory effects on mast cell degranulation and mast cell histamine release, the β-hexosaminidase release assay and the IL-4 and TNF-α secretion assay.
The results of the research showed that the EDA of both C oil and G oil was high, especially with CG oil displaying a considerably high antioxidant capacity of 92.3% (p<0.001). The results of measuring SOD like activity showed that the SOD like activity of both C oil and G oil was high, especially with CG oil displaying a considerably high SOD like activity effect of 91.0% (p<0.001). The results of measuring LPA also showed that the LPA of both C oil and G oil was high, with CG oil displaying a considerably high LPA of 97.9% (p<0.001). This indicates a high antioxidant capacity of C oil, G oil and CG oil. Regarding the inhibitory effect on the histamine release from RBL-2H3 cells, both C oil and G oil displayed an inhibitory effect in comparison with control groups and CG oil displayed a considerable synergy effect of 33.10% (p<0.01). As for the inhibitory effect on the β-hexosaminidase secretion, both C oil and G oil displayed an inhibitory effect and CG oil displayed a synergy effect of 38.39% (p<0.01). With regard to the inhibition of IL-4 and TNF-α production, both C oil and G oil displayed an inhibitory effect, and CG oil displayed synergy effects of 27.98% (p<0.05) and 22.32% (p<0.01) as to IL-4 and TNF-α, respectively.
The results of the research have confirmed that supercritical carbon dioxide extracted Chamaecyparis obtusa oil (C oil) and German chamomile oil (G oil) are worth utilizing in the alternative medicine treatment of AD and that a mixture of Chamaecyparis obtusa and German chamomile oils (CG oil) has a synergy effect. From the results of the research, such utilization is expected the clinical treatment using aromatherapy.
