ABSTRACT

Chemical Composition and Biological Activity of Commercially Available Essential Oils

The essential oils, well-known aroma compounds, have extensively been used in health care or pharmacology field. This study was studied the chemical compositions of five kinds of commercially available essential oils in Korea and their antimicrobial and antioxidant activities. The chemical compositions of oils were analysed by gas chromatography and mass-spectrometry (GC-MS), their antimicrobial activities against forty bacterial strains by tube dilution method, antioxidant activities by 1,1-diphenol-2-picrylhydrazyl (DPPH) assay and the measurement of reducing power, and the further elucidation of cell protective effect of essential oils by lactate dehydrogenase (LDH) assay.
The minimum inhibitory concentrations (MIC) were in the ranges of 0.0625-0.5% (v/v) for lemongrass; 0.0625-1.0% for manuka; 0.025-1.0% for tea tree; 0.0625-1.0% for thyme and 1-4% for ravensara. This study showed that, among the essential oils tested, lemongrass and thyme oil had broad antimicrobial spectrum against infectious bacteria. Antioxidant activities of the essential oils were determined by DPPH and reducing power. Thyme oil showed the strongest antioxidant activity among 5 oils, compared with ascorbic acid. Thyme pre-treated SH-SY5Y cells showed slightly decreased release of LDH in the concentration of 5 ㎍/㎖ solely among five oils. Thyme also attenuated somewhat rotenone-induced intra-cellular reactive oxygen species (ROS) production in cells in a concentration-dependent manner.
This study will be the report on the chemical composition of commercially available essential oils in Korea and on antimicrobial activities against gram positive or negative standard strains and multi-drug resistant (MDR) bacterial strains isolated in domestic area.
