Abstract
In the middle of searching the potential treatment drugs of stomach cancer which is one of the main cancers in Korea, this study was designed the following research to find out the effects of Euonymus alatus and Zizania latifolia applied to a variety of a folk remedy. This study was confirmed phenolic and flavonoid contents, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity to check the biological active and observed 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay, Western blot and fluorescent microscope after experimenting with extracts from Euonymus alatus and Zizania latifolia in four kinds of cancer cells (AGS, Hela, HepG2, and MCF-7) to check anticancer activity.
An extraction rate of Zizania latifolia extracts (7.3±0.1%) was twice higher than that of Euonymus alatus extracts (3.5±0.3%), but phenolic contents in Euonymus alatus extracts (277.4±7.8 GAE mg/g) was measured higher than in Zizania latifolia extracts (25.2±0.4 GAE mg/g). Also in flavonoid contents, Euonymus alatus extracts (13.1±1.1 QE mg/g) was measured higher than in Zizania latifolia extracts (6.5±0.7 QE mg/g). Euonymus alatus extracts was measured higher than Zizania latifolia extracts in antioxidants scavenging DPPH and ABTS radical, which is considered to contribute to phenolic and flavonoid contents. Euonymus alatus extracts revealed higher antioxidants than different extracts in an existing report, previewing the possibility to develop into natural antioxidants.
As a result of confirming anticancer activity of extracts from Zizania latifolia extracts and Euonymus alatus, Zizania latifolia extracts didn't have anticancer activity but Euonymus alatus extracts showed anticancer activity that peculiarly reduced only cancer cells, decreasing the survival rate of cancer cells. There was the highest anticancer activity in AGS, followed by Hela, HepG2, and MCF-7. Also when experimenting with them for 48 hr. or 72 hr., they showed anticancer activity that isn't tolerant of cancer cells, resulting in constant decrease on cell survival.
As a result of conducting Western blot after disposing of Euonymus alatus extracts from AGS, Bax, pro-apoptotic factor, was increased and Bcl-2, anti-apoptotic factor, was decreased, which made intrinsic apoptosis occur and caspase-3 activated.
Based on these results, Euonymus alatus ethanol extracts showed the possibility of effective anticancer drugs. Also these results can be an opportunity to develop Euonymus alatus anticancer drugs. Thus it can be concluded that Euonymus alatus extracts can be used in developing medicine for anticancer drugs.
