- Abstract
- Psoralea corylifolia is widely distributed over southeastern Asian countries and has been used as traditional Chinese medicine. Bavachin, one of the main constituents of Psoralea corylifolia, possess various biological activities in common such as inhibiting platelet aggregation, anti-oxidation and anti-bacterial activity. However, the anti-cancer activity of bavachin has not yet been well defined. In this study, therefore, the cytotoxic activity of bavachin and the mechanism of cell death exhibited by bavachin were examined in KB human oral cancer cells.
The cytotoxic effect of bavachin on cell growth in KB cells was examined by inhibition of cell growth (MTT assay). In addition, the cell death mechanism by bavachin was examined using DAPI staining, DNA fragmentation analysis and immunoblotting.
1. Treatment of bavachin in KB cells induced the inhibition of cell growth in a concentration- and a time-dependent manner as determined by MTT assay.
2. Treatment of bavachin induced the nuclear condensation in KB cells.
3. Treatment of bavachin induced the DNA fragmentation in KB cells.
4. The proteolytic processing of caspase-3, -7, -8 and -9 was increased by bavachin treatment in KB cells.
These results suggest that bavachin can induce the suppression of cell growth and cell apoptosis in KB human oral cancer cells, and therefore, that it may have potential properties for anti-cancer drug discovery.
